THE RATIONALE

Why combine retatrutide and vitamin B1?

Retatrutide produces profound metabolic and appetite effects. Panacea Bio Chem developed the Rethiamine concept in response to the emerging scientific concern that powerful appetite suppression, reduced dietary intake and gastrointestinal intolerance may leave some individuals nutritionally vulnerable.

Rethiamine does not assume that retatrutide directly consumes or chemically depletes thiamine. That mechanism has not been established.

The scientifically defensible framing is retatrutide-associated nutritional vulnerability: potent appetite suppression, reduced dietary intake, gastrointestinal intolerance and rapid weight reduction may create nutritional vulnerability in some individuals. Panacea Bio Chem is investigating whether integrating thiamine into a once-weekly retatrutide formulation could provide a more coherent pharmaceutical research architecture.

What thiamine does — the established biology

  • Thiamine is required for cellular energy metabolism, including carbohydrate and amino-acid metabolism.
  • It is converted into thiamine diphosphate, the coenzyme form used by thiamine-dependent enzyme systems.
  • It participates in nervous-system function.
  • Humans maintain limited thiamine reserves, so status depends on continuing intake.
  • Severe restriction of food intake and prolonged vomiting can contribute to thiamine deficiency.

What these support: the physiological background for the B1 component. What they do not show: thiamine co-formulated with any incretin drug, or dosing in combination injectable products.

The nutritional-vulnerability concern

Current research has raised concern about nutritional deficiencies during treatment with potent appetite-suppressing incretin therapies, particularly in the presence of severe dietary restriction, nausea or vomiting. A 2026 Nutrients review argues for structured nutritional management — including micronutrient monitoring — during incretin-based therapy, whose gastrointestinal adverse events can compromise intake and adherence (Pardali et al., PMID 41901137 ↗). What this supports: the relevance of nutritional monitoring during substantial pharmacologically driven weight loss. What it does not show: a quantified thiamine deficiency risk on GLP-1/GIP/glucagon agonists, or any tested supplementation strategy.

Direct retatrutide-induced biochemical depletion of thiamine has not been established. Rethiamine™ is Panacea Bio Chem’s proprietary formulation response to this emerging research question and remains subject to analytical, pharmaceutical, toxicological and clinical evaluation.

Why inside the same weekly shot?

Retatrutide supplies weekly pharmacological exposure. The Rethiamine hypothesis asks whether a thiamine component can be integrated into the same weekly administration event — one coherent architecture rather than a separate supplement regimen. Whether a weekly thiamine component can meaningfully support thiamine status is an open investigation, not a claim; the proposed 20 mg inclusion is a formulation-development strength, not an established clinical requirement.
